Detailed methodology and results are described in following publication:<\/strong> Briefly (excerpted from above):<\/strong> On day 11 of the experiment, prior to the commencement of\u00a0settlement and metamorphosis, larvae were transferred into\u00a0new glass \"substrate\"\u00a0jars.\u00a0The surface of each new jar base was abraded with\u00a0sandpaper to encourage larval settlement.\u00a0The underside of\u00a0each new jar base was scored to facilitate separation into four\u00a0tiles after larvae had metamorphosed into benthic juveniles.\u00a0These tiles were suitable for outplanting juvenile oysters to\u00a0the field.<\/p>\n Seawater chemistry:<\/em> Metamorphosis and field outplants:<\/em> A shoreline region half-way along the Tomales Bay estuary\u00a0was selected for the two replicate field sites (38\u00b0 09'\u00a001.01\"N,\u00a0122\u00b0 53'\u00a019.19\"W). The replicate sites were 40 m apart in the\u00a0alongshore direction, and were similar in substrate type, solar\u00a0exposure, and bottom slope.\u00a0At each replicate site, six PVC \u2018T\u2019\u00a0stakes, placed 0.5 m apart and each holding two tiles, were\u00a0driven into the substrate such that the tiles were situated at\u00a0either 0 or 0.3 m above MLLW (i.e., 'low'\u00a0and 'mid'\u00a0shore\u00a0levels, respectively). Tiles deployed on four of the six stakes\u00a0per site and shore level were outfitted with temperature loggers\u00a0(iButton, Maxim, Sunnyvale, CA, USA).\u00a0A YSI 6600V2 multi-parameter instrument was used to measure\u00a0temperature, salinity, and pH(NBS) at each of the outplant\u00a0sites approximately weekly up to day 52\u00a0postsettlement, and\u00a0then monthly up to day 127 postsettlement.<\/p>\n Juvenile survival on each tile was calculated as the percentage\u00a0of initial juveniles that were alive on each sampling date\u00a0(day 6, 13, 27, 127 postsettlement).\u00a0Each tile was examined for live juvenile oysters under a\u00a0dissecting microscope (Leica M125 with DC290 camera) at BML.\u00a0Juvenile growth rates were estimated from photographs of\u00a0juveniles randomly sampled on each tile using a random\u00a0number table to select squares on the gridded tile.\u00a0Juvenile growth rates\u00a0were calculated on days 6, 13, and 27 postsettlement\u00a0as the\u00a0change in the total projected area of the shell between the\u00a0sample date and when the larvae metamorphosed, divided by the intervening number of days (mm2 per day). Larval culturing:<\/em>
\nAdult Olympia oysters (n = 105) were collected in September\u00a02010 from Tomales Bay, California (38\u00b0 06'\u00a057.01\"N, 122\u00b0 51'\u00a014.39\"\u00a0W) and transported to\u00a0Bodega Marine Laboratory (BML). Adults were\u00a0cleaned of epiphytes and distributed evenly among three 100 L\u00a0cylinders.\u00a0Adults\u00a0were fed microalgal food to encourage\u00a0larval release.\u00a0After 72 h, larvae were released in one of\u00a0the cylinders, and subsets of these individuals were distributed\u00a0by pipette among 4.5 L\u00a0glass jars (n = 1000\/jar) used for\u00a0culturing.\u00a0Larvae were\u00a0reared through their entire planktonic duration in a\u00a0culturing facility at BML.\u00a0The target seawater pCO2 concentration in larval treatment\u00a0cultures was 1000 uatm. The accompanying control seawater\u00a0pCO2 target was 400 uatm.\u00a0After filling culture\u00a0jars with carboy water, pCO2 concentrations were maintained\u00a0in the jar seawater by pumping the same CO2 gas mixtures\u00a0into sealed air spaces above the free surfaces of the seawater\u00a0in the culture jars (\"headspaces\") shared by 6 replicate\u00a0jars per pCO2 concentration.\u00a0There were three headspaces for each pCO2 concentration,\u00a0and 6 jars associated with each headspace (n = 2 pCO2\u00a0concentrations x\u00a03 headspaces x\u00a06 replicate jars = 36 jars).<\/p>\n
\nSeawater pH(NBS) and temperature were quantified\u00a0using a potentiometric pH\/temperature meter (Accumet\u00a0Excel XL60), salinity was determined using a YSI 6600V2\u00a0multi-parameter instrument, and total alkalinity (TA, umol\u00a0per kg of seawater) was measured using automated Gran titration\u00a0with duplicates (Metrohm 809). A subset of samples was\u00a0analyzed for dissolved inorganic carbon (DIC, umol per kg of\u00a0seawater) at the University of Georgia\u2019s infrared CO2 analysis\u00a0facility (Cai & Wang, 1998). Both TA and DIC measurements were standardized using\u00a0certified reference material from A. Dickson at Scripps Institute\u00a0of Oceanography (La Jolla, California). Calcite and aragonite\u00a0saturation states (\u03a9calcite, \u03a9aragonite) and seawater pCO2\u00a0were calculated using the carbonate system analysis software,\u00a0CO2SYS (Lewis & Wallace, 1998).<\/p>\n
\nSettlement of larvae and metamorphosis into benthic juveniles\u00a0was assessed daily starting at day 11 when larvae were transferred\u00a0into substrate jars.\u00a0Each jar\u00a0base was divided into four 50 cm2 tiles for outplanting. Tiles from a given pCO2 concentration with similar\u00a0juvenile densities were arranged into four groups, each\u00a0composed of six replicate tiles. The four groups were then randomly\u00a0assigned to one of the two field sites and one of the two\u00a0shore levels.\u00a0The overall design was: n = 2 larval\u00a0pCO2 concentrations x\u00a02 sites x\u00a0shore levels x\u00a06 tiles = 48\u00a0tiles.
